anti-human LAT1, L-type amino acid transporter 1

Background:
Mammalian cells have several amino acid transport systems. These systems have been defined by both activity and specific genes.  L-type amino acid transporter 1 (LAT1) is a Na + -independent neutral amino acid transporter and essential for the transporter of large neutral amino acid such as leucine, isoleucine, and valine through the plasma membrane. LAT1 has been proposed to be one of the major nutrient transport systems at the blood-brain barrier. Drugs such as L-Dopa are transported by LAT1.  LAT1 is thought to be up-regulated to support the high protein synthesis for cell growth in some tumor cell lines.  LAT1 has also been referred to as Large neutral amino acids transporter small subunit 1, 4F2 light chain (4F2 LC or 4F2LC). CD98 light chain, Integral membrane protein E16, and hLAT1.

Antibody Description:
Rabbits were immunized with the KLH conjugated synthetic peptide C-RHRKPEL representing a portion of the sequence of human LAT1. The serum was pooled in equal volumes from both rabbits for two production bleeds one week apart. The antibody is provided as Protein G purified IgG in PBS, pH 7.4 with no preservatives to preclude any potential chemical interference in use of the product. The antibody concentration was determined by extinction coefficient where an O.D. reading of 1.4 at 280 nm equals 1.0 mg/ml. The purified antibody should be aliquoted and stored frozen to avoid numerous freeze-thaw cycles.

Application:
For immunohistochemistry this rabbit antibody to human LAT1 compares well with a commercially available monoclonal. A dilution of 5 ug/mL in conjunction with the LSAB2 kit from DakoCytomation is optimal.
