Sheep anti-Bromodeoxyuridine (BrdU)

Description: An anti-BrdU antibody is valuable for proliferation studies. BrdU is used for nucleotide substitution to replace thymidine with uridine in the DNA structure of dividing cells of various types. This antibody has a long history of use in research and has been widely published.

Specifications: For initial characterization product was tested using immunoprecipitation against 5-Methyl Cytosine (5-MeC) and BrdU along with no antigen control and assayed by A 405 nm spectrophotometry. At a concentration of 25 µg/mL, this product demonstrates 8 fold higher reactivity with 5-MeC versus BrdU. This product titers out at 50 ng/mL. For best results, use the antibody at a concentration of 25 to 100 µg/mL. Nearly complete immununoprecipitation was obtained at a concentrations of 100 µg/mL. Anti-BrdU is used for for staining of bromodeoxyuridine incorporated into DNA of replicating cells. When utilized in 10 µg/mL concentration, the product is comparable to commercially available monoclonal antibodies commonly used for the same purpose. This sheep reagent is useful for multiple labelling experiments. Denature DNA sample first so that bases are accessable to BrdU antibody. (Protocol available upon request and through this link).

Formulation: 250 µg of purified IgG in PBS with no preservatives

Frozen and paraffin sections, starting dilution ~ 10 µg/mL. First use a fixation step in cold methanol for 30 minutes followed by immersion in 0.007 M NaOH for 10-15 seconds for BrdU staining with the simultaneous detection of nuclear cytoplasmic and membrane components as well as preservation of morphological detail.
