Sheep anti-Angiotensin II type 1 (AT(1)) Receptor

Background: Angiotensin II was originally described as a vasoactive peptide that regulates blood pressure. However, it has become clear that the renin-angiotensin system has a broad distribution in a wide variety of tissues and plays a role not only in regulation of blood pressure but also in salt and water metabolism. It has also been found to play a role in a wide variety of central nervous system functions. Two major subtypes of angiotensin II receptors have been cloned. These are known as the AT-1 and AT-2 receptors. The AT-1 receptor has been the major focus of study since it appears to mediate all of the vasoactive properties of angiotensin II. (It should be noted that there are pharmacological data supporting the existence of at least two additional angiotensin II receptors.) In addition, angiotensin II, which is an 8-amino acid peptide (1-8), can be metabolized by a variety of amino-peptidases to shorter peptide fragments (most notably the 1-7 and 3-8 forms of the peptide). These bind to additional angiotensin receptors in the vasculature and other organs that are yet to be defined.

Antibody Description:
Sheep were immunized with GST-fusion protein Angiotensin II, Type-1 receptor, amino acids 297-359. This amino acid sequence is thought to be responsible for receptor-associated kinase activity. This antibody will not effect binding activity of the receptor. This antibody is offered either as serum or Protein G purified IgG. The purified antibody is provided at a concentration >1.0 mg/mL in PBS, pH 7.4. The antiserum is highly immunoreactive and is specific to the AT-1 receptor and can be diluted to >1:10^8 before immunoreactivity is lost in direct EIA. The antiserum has also been used in Western Blot (WB) at greater than 1:2000 using myocardial membrane homogenates. WB also tested on mouse proximal convoluted tubules and rat kidney samples.

Storage: Aliquot and store at -20° C or colder. Once thawed store at 4° C with the addition of 0.1% sodium azide. Avoid repeated freeze-thaw cycles.
